Distinctive Characteristic

Form developed to support brain magnesium

What Is Magnesium L-Threonate?

Magtein is magnesium bound to L-threonate, a form developed specifically to support brain magnesium levels. Magnesium is a cofactor in hundreds of enzymatic reactions, but most forms — oxide, citrate, glycinate — are not designed around brain delivery. Research in animal models suggested L-threonate could support elevation of magnesium in the cerebrospinal fluid, motivating human studies on cognition, memory, working memory, sleep, and daytime function. Unlike magnesium oxide (high elemental magnesium but poorly absorbed) or magnesium citrate (well absorbed but not brain-targeted), L-threonate is positioned for neurological and cognitive applications. Wellnex selects magnesium L-threonate when the formula objective is cognitive, sleep, or stress support rather than general magnesium repletion.

Magnesium L-Threonate vs Other Forms of Magnesium

Magnesium forms differ in bioavailability, tolerability, and intended application. Magnesium oxide has high elemental magnesium but low absorption. Magnesium citrate is well absorbed and commonly used for repletion and bowel regularity. Magnesium glycinate (bisglycinate) is well tolerated and used for relaxation and sleep support. Magnesium L-threonate was developed around brain delivery: preclinical research suggested it raises CSF magnesium, and human trials have examined cognition, memory, and sleep outcomes. No single form is superior for every application; form selection depends on the clinical objective. For neurological and cognitive formulas, L-threonate is the form with the most targeted research base.

Research Highlights

Evidence Distinction

Research labeled “Branded Ingredient Study” used the named branded material. Research labeled “Ingredient-Level Research” studied the nutrient or ingredient class generally and is informative without implying the same exact material was used. We do not imply a study used a branded ingredient unless the publication identifies it.

Population:Healthy adults 18–65 y
Participants:109
Duration:30 days
Dose:Magtein®PS (Mg L-threonate + phosphatidylserine + vitamins C & D) 2 g/day vs placebo
Primary Area:Clinical Memory Test (memory & cognition)

Key Finding

Magtein®PS significantly improved all Clinical Memory Test subcategories and the overall memory quotient versus placebo, with greater improvement in older participants.

The effects of magnesium L-threonate (Magtein®) on cognitive performance, cognitive age, sleep quality, and selected physiological indicators in adults: a randomized controlled trial

Lopresti AL, Smith SJ

Frontiers in Nutrition, 2025

In a 6-week RCT, Magtein® improved overall cognition, working memory, reaction time, and autonomic balance (HR/HRV) in adults with dissatisfied sleep.

Population:Adults 18–45 y with self-reported dissatisfied sleep
Participants:100
Duration:6 weeks
Dose:Magtein® 2 g/day vs placebo
Primary Area:Cognition (NIH Toolbox), sleep, heart rate, HRV

Key Finding

Magtein® improved overall cognition (NIH total composite), working/episodic memory, reaction time, and autonomic balance (lower resting HR, higher HRV), and reduced estimated brain cognitive age; subjective sleep-related impairment improved in a subset.

Frequently Asked Questions

What is magnesium L-threonate?

Magnesium bound to L-threonate, a form developed specifically to support brain magnesium levels.

Does magnesium L-threonate cross the blood-brain barrier?

Preclinical research suggested L-threonate supports elevation of magnesium in cerebrospinal fluid; the clinical significance is an active area of study.

What is the difference between magnesium glycinate and magnesium L-threonate?

Magnesium glycinate emphasizes tolerability and general relaxation; magnesium L-threonate was developed for cognitive and brain-focused applications.

What is Magtein?

Magtein is a branded magnesium L-threonate ingredient used in cognitive and sleep-focused formulas.

What is the best magnesium for brain health?

No form is best overall. Magnesium L-threonate has the most brain-targeted research; selection depends on the formula goal.
